As a Clinician Scientist in the Rieger lab Dr. Daniel Fischer investigates B-cell precursor acute lymphoblastic leukemia (BCP-ALL) and its mechanisms of CNS infiltration. Using a lentiviral barcoding system and long-term ALL cell cultures derived directly from patients, leukemic cells are labeled and tracked in vivo at the clonal and single-cell level. The analyses reveal significant clonal diversity between organs and unique characteristics of leukemia cells in the brain, including activation of genes relevant for migration and metastasis. These findings provide new insights into the development of therapy-resistant relapses and could enable more targeted treatment strategies in cases of CNS involvement.
